ABSTRACT
Taufan Febri Yuanto, The Effectiveness of Cytocinine Hormone for the Growth
and Development of Melon’s Plantlet Axillary Sprout (Cucumis melo L.),
advised by Netty Ermawati, SP. Ph.D and Ir. Hari Prasetyo, MP.
This research aims to identify the type of cytocinine hormon and its best
concentrate on triggering the growth of Melon’s explants through in vitro method.
The research was conducted in June 2013 to March 2014 at BIOSAIN Laboratory
of Politeknik Negeri Jember. The design used was a completely–randomized
design Non–Factorial that is the variety of cytocinine levels were used as
treatment, they were ; Kinetin (0,1 ppm. 0,5 ppm. 1 ppm), BAP (0,5 ppm. 1 ppm. 2
ppm), Coconut water (5%, 15%, 25%) added by 0,2 ppm NAA. All treatments
were used MS + 0,2 ppm NAA as a based media, and MS without NAA used as a
control. Every treatment was repeated for three times. This research was divided
into two parameters, the main parameter amd the additional parameter. The main
parameter consists of the quantities of sprouts, roots, leaves, and segments. And
the additional parameter consists of the emanating celerity of calluses, roots, and
sprouts. The best growth and development response on twelefth week was showed
by MS without NAA 0,2 ppm within the average of sprout quantity reached out
into 1,00 sprout. The quantities was 29,33 for the leaves, 15,33 for the segments,
and 5,00 for the roots. As for the secondary parameter, MS treatment + Kinetin
0,5 ppm + 0,2 ppm NAA showed the highest celerity average in emanating the
callus reached out into 26,67 days. MS without NAA 0,2 ppm showed the highest
average of the celerity parameter in emanating sprouts reached out into 43,67
days. While MS treatment + Kinetin 0,1 ppm + NAA 0,2 ppm showed the best
response of celerity parameter in emanating roots with the highest average
reached out into 43,67 days. The result of this research found that using some
cytocinine Hormone isn’t effective to growth and development of melon’s planlet
axillary sprout.
