ABSTRACT
Orchids are known having a compelling fascination, it causes the market demand of Cattleya orchid will also increase. Orchids are difficult to breed than through tissue culture. Every explant from different commudities have different suitabillity of the media. In previous study, Murashige and Skoog (MS) was found better media compared companed to Vacin and Went (VW) for orchid germination. The purpose of this study is to find the appropriate media for subculture of cattleya orchid. The study was conducted from January to July 2014 at Tissue Culture Laboratory of Agricultural Production Departement State Polytechnic of Jember. The study was conducted by using the completely randomized design (CRD) Non Factorial using 8 treatments, repeated 3 times. The medium treatments were divided into 8, namely M1 = MS medium, M2 = MS + Coconut Water 75 ml/l medium, M3 = MS + Coconut Water 75 ml/l + Potatoes 100 g/l medium, M4 = MS + Potato 100 g/l medium, M5 = VW medium, M6 = VW + Coconut Water 75 ml/l medium, M7 = VW + Coconut Water 75 ml/l + Potatoes 100 g/l medium and M8 = VW + Potato 100 g/l medium. The parameter observations were the rate of root emergence, number of roots, number of leaves and number of shoots. The result shown that the rate of root emergence in M1 treatment shown the best result by produced the namber of roots 3,28 in 60 day after growing (DAG), followed by M3 treatment produced the roots after 17,72 DAG. The highest amounts of root was shown in M1 treatment in 90 DAG produced 3,61 roots and the amount of shoots was shown by M3 treatment 1n 120 DAG by produced 0,67 of shoots.
