ABSTRACT

The iles-iles plant (AmorphophallusmuelleriBlume) of the Araceaetribe, is a tuber-producing plant that is included in the tubers and is called iles-iles tubers. Propagation and growth of iles-iles takes a very long time so it needs to be multiplication by using tissue culture techniques. This study aims to determine the effectiveness of growth substances grown Thidiazuron (TDZ) is right in regenerating callus iles-iles. This research was conducted for 4 months since February 2018 until May 2018. The research place is in Tissue Culture Laboratory, State Polytechnic of Jember. This research was conducted using non-factorial completely randomized design (CRD) with 5 treatments and 4 replications including 5 Thidiazuron concentration levels of 0.1 ppm, 0.2 ppm, 0.3 ppm, 0.4 ppm and 0.5 ppm. Data were analyzed using Analysis of Variance and then tested continued using DMRT with 5% and 1% level. The result of research showed that thidiazuron growth regulator in MS media had no significant effect on all parameters of study including callus formation, budding buds, root formation, number of buds, shoot height, and root number.

Efektivitas ZPT Thidiazuron Terhadap Pertumbuhan Iles-iles (Amorphophalus muerelli Blume) secara In Vitro

ABSTRAK

Tanaman iles-iles (Amorphophallus muelleri Blume) dari suku Araceae, adalah tanaman penghasil umbi yang termasuk dalam umbi batang dan disebut umbi iles-iles. Perbanyakan dan pertumbuhan iles-iles membutuhkan waktu yang sangat lama sehingga perlu dilakukan perbanyakan dengan menggunakan teknik kultur jaringan. Penelitian ini bertujuan untuk mengetahui efektifitas zat pengatuh tumbuh Thidiazuron (TDZ) yang tepat dalam pertumbuhan iles-iles. Penelitian ini dilaksanakan selama 4 bulan sejak bulan Februari 2018 sampai bulan Mei 2018. Tempat penelitian berada di Laboratorium Kultur Jaringan, Politeknik Negeri Jember. Penelitian ini dilaksanakan menggunakan rancangan acak lengkap (RAL) non faktorial dengan 5 perlakuan dan 4 ulangan meliputi 5 level konsentrasi Thidiazuron yaitu 0,1 ppm, 0,2 ppm, 0,3 ppm, 0,4 ppm, dan 0,5 ppm.Data dianalisa menggunakan Analysis of Variance lalu diuji lanjut menggunakan DMRT dengan taraf 5% dan 1%. Hasil penelitian menunjukkan bahwa pemberian zat pengatur tumbuh Thidiazuron pada media MS berpengaruh tidak nyata (non significant) pada seluruh parameter penelitian yang meliputi Kedinian terbentuknya kalus, kedianian terbentuknya tunas, kedinian terbentuknya akar, jumlah tunas, tinggi tunas, dan jumlah akar.
