Abstract
Anther culture has been an effective method to reducing breeding time. This method allows fast achievement to obtain a homozygous line in one generation of the desired plant. The success of anther culture is strongly influenced by the composition of planting media, in addition to genetic factors. The purpose of this study was to determine the most effective types of amino acids and types of rice varieties in anther culture development. This research was conducted at the Laboratory Center of Biosciences, State Polytechnic of Jember, using Faktorial Randomized Block Design with two factors. First factor was rice varieties (MSP9 (V1), Gorontalo (V2), and Nagdong (V3)). The second factor was additional of amino acids in the basic medium (Casein Hidrolisat (A1), Proline (A2), Glutamine (A3)). The observation parameters including level browning of anther, speed of anther form callus, number of anther form callus, number of callus form planlet. The result showed that amino acid treatment on some varieties have significant effect on the development of rice anther. The result indicated medium with proline was the most efective formulation for rice anther culture, it produce 32,8 embriogenic callus, and 0,6 callus regenerates. The best varietes was Gorontalo producing 30,8 embriogenic callus, and 0,6 callus regenerates. The best treatment interaction was A2V2 producing 35,5 embriogenic callus, and 1,0 callus regenerates.
